Cohan starred as Charlotte Higginson in the sequel to Van Wilder: The Rise of Taj in 2006. This was after her film debut Casanova as Sister Beatrice. In addition, the 2008 film Float was her next role. She was cast in Death Race 2: Frankenstein in February 2010. Alongside Sean Bean and Danny Trejo. Moreover, she played the main character Greta in the supernatural-horror film, The Boy. This was released in 2016. She also appeared as Martha Wayne in Batman v Superman. Cohan’s first major supporting role was in 2007 for season three of Supernatural as Bela Talbot. The role of a thief who procures valuable supernatural objects. Then sells them in the supernatural world to very rich, very powerful people. Moreover, she appeared in six episodes of the series. In season three of Time Is On My Side, was her last in the penultimate episode.
In addition, in The Vampire Diaries, she played a recurring character of Rose, a 560-year-old vampire. She joined the television series, Chuck, in 2011 in a recurring role-playing Vivian McArthur Volkoff. She was cast in her most notable role as Maggie Greene on The Walking Dead. The AMC post-apocalyptic horror television series and the comic book series have the same name. In season two she appeared as a recurring character. However, at the start of season three, she became a series regular. It was the highest-rated series in cable television history in 2015. She completed her contract obligations as a series regular at the conclusion of the eighth season of The Walking Dead. However, due to a payment dispute, she did not reach an agreement to sign on for season nine as a main cast member. She began making herself available for TV pilots. She departed the series in 2018. This was after she had reached an agreement to appear in five episodes in the first half of season nine of The Walking Dead.
In addition, in the ABC action comedy-drama, Whiskey Cavalier, she was cast as CIA operative Francesca “Frankie” Trowbridge. This was in the midst of The Walking Dead contract negotiations with AMC. Whiskey Cavalier premiered on February 24, 2019, and was picked up to series in May 2018. However, it was not renewed for a second series by AMC. It was reported in October 2019 that she would return to The Walking Dead as a series regular in the series Eleventh and final season.
